Quantitation of HCV
Special Instructions
Submit a separate frozen specimen for each frozen test ordered. Values obtained with different assay methods should not be used interchangeably in serial testing. It is recommended that only one assay method be used consistently to monitor each patient's course of therapy. This procedure does not provide serial monitoring; it is intended for one-time use only. If serial monitoring is required, please use the serial monitoring number 550033 to order.
Specimen
Serum or plasma, frozen
Volume
2 mL
Minimum Volume
1 mL
Container
Red-top tube, gel-barrier tube, plasma preparation tube (PPT™), yellow-top (ACD) tube, or lavender-top (EDTA) tube. Do not use green-top heparin tube.
Collection
Collect whole blood in red-top tube, gel-barrier tube, PPT™, yellow-top (ACD) tube, or lavender-top (EDTA) tube. Do not use green-top (heparin) tubes. Centrifuge specimen within 6 hours of collection. Remove plasma or serum, transfer specimen to polypropylene screw-cap tube (not “pop top” or “snap cap”), and freeze. Specimens drawn into PPT™ need to be centrifuged within 6 hours and frozen. Ship frozen. To avoid delays in turnaround time when requesting multiple tests on frozen samples, please submit separate frozen specimens for each test requested.
Storage Instructions
Freeze
Causes for Rejection
Hemolysis; green-top (heparin) tube; specimen not frozen on date of collection; PPT™ not centrifuged; specimen received in “pop-top” or “snap-cap” tube; specimen shipped at room temperature
Use
Determine the number of international units (IU) of hepatitis C virus (HCV) RNA per milliliter serum or plasma in known HCV-positive patients
Limitations
The HCV QuantaSure™ Plus assay has a quantitative range of 10 IU/mL to 100,000,000 IU/mL.
Methodology
TaqMan™ polymerase chain reaction (PCR) amplification and detection
