Special Instructions

Indicate pertinent clinical diagnosis, previous cytogenetics studies and gestation age on the test request form. There may be an additional charge for formalin positive tissue samples requiring embedding and pathology. Note: This test number is used for tracking FISH specimens to the testing laboratory. Bill codes will be added when testing is complete based on the processes performed in the laboratory. Call Customer Service at 800-345-4363 for a list of applicable test codes.

Expected Turnaround Time

7 - 10 days

Specimen Requirements

Specimen

10% neutral-buffered formalin or formaldehyde fixed fetal or paraffin-embedded tissue or cut paraffin slides.

Volume

4x4x4 mm formalin fixed fetal tissue, one paraffin block or nine cut paraffin slides (cut at 4 to 5 microns)

Minimum Volume

2x2x2 mm formalin-fixed fetal tissue, one paraffin block or five cut paraffin slides (cut at 4 to 5 microns). Note: Minimum volumes may not allow for repeat testing.

Container

Formalin-fixed fetal tissue samples: Sterile container. Do not use urine containers for shipping.

Paraffin blocks: Paraffin block transport pouch

Paraffin slides: Plastic slide container

Collection

Aseptically obtain a small piece of tissue with subepidermal layers. Best results are obtained from placental villi, which remain viable much longer than fetal tissue. This is especially true in stillborn cases in which both cord blood and tissue samples are often not viable. The sample should be fixed in 10% neutral-buffered formalin or formaldehyde then paraffin-embedded or shipped to the testing laboratory in a sterile container. Transport to the testing laboratory at room temperature is acceptable. Do not allow the sample to overheat or freeze. Use a cool pack or LabCorp transport kit if necessary.

Storage Instructions

Maintain specimen at room temperature and transport to the cytogenetics laboratory immediately. Do not freeze.

Causes for Rejection

Stained paraffin blocks or slides; decalcified blocks or slides; broken slides; absence of fetal tissue; low volume or quantity not sufficient for analysis

Test Details

Use

Rapid identification of common aneuploidies (specific for X, Y, 13, 16, 18, 21, 22) as the cause of miscarriage.

Limitations

Molecular mutations below the resolution of FISH will not be detected. Will not detect regions not targeted by FISH probes in this panel.

This test was developed, and its performance characteristics determined, by LabCorp. It has not been cleared or approved by the US Food and Drug Administration (FDA).

Methodology

Fluorescence in situ hybridization (FISH)
